ABSTRACT

Heme oxygenase (HO) -1 is the most bioactive HO type. It reduces tissue damage by exerting antioxidant, anti-inflammatory, and cytoprotective effects during ischemia-reperfusion injury. It is considered to be a new direction for the treatment of ischemia-reperfusion injury. This article reviews the role of HO-1 in ischemia-reperfusion injury and its possible mechanisms.

ABSTRACT

Objective To explore the expression level and possible reasons of Tau protein in Beagle dog model of ischemic cerebral white matter(WM) demylination.Methods Sixteen adult Beagle dogs were divided into the sham operation group(A) and observation group B,C and D according to the completely random mumber table method,4 cases in each group.Different.degrees of cerebral ischemia animal models were established by 4-vessel occlusion(4-VO) method.The bilateral ventricle edge white matter (WM) was selected.The oligodendrocyte precursors(OPCs) were labeled by NG2.The mature oligodendrocytes(Ols) were labeled by CNPase.Tau,NG2 and CNPase were detected by using the immunohistochemical method.The expression level was quantified by the mean optical value.The correlation among Tau,NG2 and CNGase was analyzed by adopting the Pearson linear correlation analysis.Results The HE staining showed obvious changes of WM demylination after chronic cerebral ischemic.The scores after LFB staining in the group A,B,C and D were(0.75 ± 0.71) points,(1.38 ± 0.06) points,(1.63 ± 0.52) points and (1.88 ± 0.64)points.Compared with the group A,the scores in the group B,C and D were much higher(P<0.05).Compared with the group A,the expression levels of Tau protein and NG2 were significantly increased(P<0.05),while the CNPase expression level was significantly decreased(P<0.05).The Pearson correlation analysis showed that Tau expression level was positively correlated with the NG2 expression level(r=0.277,P=0.006);Tau and NG2 were negatively correlated with the CNPase level(r=-0.303,-0.402,P=0.003,0.001).Conclusion The increase of Tau expression in Beagle dog model of ischemic cerebral WM demylination may be related to the differentiation dysmaturity of OPCs.

ABSTRACT

Toll-like receptor 2 (TLR2) is the most w idely expressed receptor in the Toll-like receptor family, and is also an important pattern recognition receptor of the innate immune system to pathogenic microorganisms. In addition, TLR2 can also identify endogenous risk signals and participate in non -pathogenic microbial inflammatory reaction in ischemic injury. The role of TLR2 and its signal transduction in ischemic cerebral w hite matter lesions have received more and more attention. This article reviews the relationship between TLR2 and ischemic cerebral white matter lesion.

ABSTRACT

Magnesium alloys have been a hotspot in the field of implanted medical devices due to their biodegradable absorbability, excellent mechanical properties and good biocompatibility.The reduction in their rapid corrosion rates becomes the key to the application of implant medical device materials.In this paper, the latest research progress and the existing problems of magnesium alloys as the material for implantation of medical devices in the fracture internal fixation, bone tissue porous scaffold, and cardiovascular stent are reviewed.Improving corrosion resistant of magnesium alloys by means of alloying, improving purity, surface modification,rapid solidification, deformation processing, non crystallization and preparation of nano alloy technology in body fluid are expounded, and research direction and application prospect of magnesium alloys in the field of implanted medical devices are also expected.

ABSTRACT

Objective To investigate the relationship between serum homocysteine (Hcy) and large artery atherosclerosis (LAA).Medods The consecutive stroke patients with LAA and small artery occlusion (SAO) were enrolled.The demographics,baseline clinical data,and laboratory test results were compared.Multivariate logistic regression analysis was used to identify the independent risk factors for LAA.Spearman correlation analysis was used to analyze the correlation between serum Hcy concentration and carotid plaque scores.Results A total of 117 patients with LAA and SAO were enrolled,42 were in the LAA group and 75 were in the SAO group.The plasma Hcy levels (median [interquartile range] 15.35 [12.20-20.43] μmol/Lvs.12.40 [10.40-15.00] μmoFL; Z=-2.540,P=0.011),the carotid plaque scores (6.30 ±4.11 mm vs.4.19 ±2.84 mm; t =2.952,P=0.004),and the proportion of plasma Hcy level at tertile groups of the patients (x2 =10.697,P =0.005) of the LAA plasma group were all significantly higher than those of the SAO group.Multivariate logistic regression analysis showed that the plasma Hcy concentration was in the third tertile (> 15.3 μmol/L),and it was an independent risk factor for LAA (odds ratio 4.739,95% confidence interval 1.696-13.241; P =0.003).Moreover,there was a significant positive correlation between the serum Hcy concentration and the carotid plaque scores (r =0.245,P =0.008).Conclusions The plasma Hcy levels were positively correlated with the severity of carotid atherosclerosis.The Hcy > 15.3 μmol/L was an independent risk factor for LAA.

ABSTRACT

The collagen I was made with the dialysis method of enzymolysising the pig skin, and the static deposition in vitro of calcium phosphate was comparative studied by X-ray diffraction (XRD) and infrared spectroscopy (FTIR) under the condition of pH7. 4, Ca/P 1.67 and whether adding the collagen I into the system. Then the chemical composition of the sedimentary product and the diversification of the collagen I 's IR and Raman spectra (RS) before and after the mineralization were analyzed. The results showed that,under the physiological pH condition that there was not any collagen I, though Ca/P reached up to 1.67, the sedimentary product was CaHPO4 x 2H2O yet, however, after adding collagen I into the system, the bone-like apatite was deposited, which proved that collagen I indeed had the effects on the inducing of the bone-like apatite's mineralization in vitro; there was obviously mutual coordination action between collagen I and its mineralization product--bone-like apatite, which caused that amide peak I red-shifted, amide peak II and amide peak III decreased significantly or disappeared on the IR of collagen I, which maybe was the mechanism that how collagen I induced the depositing of the bone-like apatite.

ABSTRACT

Bone cement samples were made of tetracalcium phosphate (TTCP) and monocalcium phosphate monohydrate(MCPM) powder (Ca/P = 1.67) by using water and 5.24 mg/ml of self-made type I collagen sol as hardening liquid with the solid-liquid ratio of 3:1, their setting time and compressive strength were tested. The results showed that: the compressive strength of TTCP/MCPM bone cement containing collagen could increase from 17.8 +/- 1.9 MPa to 22.7 +/- 1.6 MPa, but its setting time hasn't been significantly affected; the compressive strength of both samples immersed in simulated body fluid (SBF) could increase, and the growth rate of the sample containing collagen increased especially; both samples immersed in SBF for 4d and 14d, whose compressive strength could increase to 31.8 +/- 3.9 MPa (collagen)/19.5 +/- 1.3 MPa and 38.1 +/- 3.1 MPa (collagen)/21.9 +/- 2.2 MPa. According to the IR analysis before and after the collagen was mineralized, it showed that: after the collagen was mineralized, the characteristic peaks of the collagen's amide I band showed red-shift, while the amide II band and the amide III band nearly disappeared, suggesting that chemical action occurred between the collagen and hydroxyapatite (HA), which should be the basis of the enhancement on the TTCP/MCPM bone cement caused by collagen; while according to the SEM and XRD patterns of the sample surface before and after the samples were immersed in SBF, it showed that: the immersion in SBF changed brushite (DCPD) into HA, at the same time, large number of new HA deposited, making the samples' surface more dense and smooth. It was not only the enhancement mechanism of immersion in SBF, but also showed the coagulating and hardening process of TTCP/MCPM bone cement was that: the DCPD was generated firstly, then it changed into HA.

ABSTRACT

A large number of laboratory and clinical researches have indicated that the occurrence of restenosis after percutaneous transluminal angioplasty is mainly related to the following factors: the inflammatory reaction, the injury of vascular wall, the aggregation of platelet, the formation of thrombus, the hyperplasia of vascular endotheliocyte, etc. The corresponding parameters related to the above mentioned factors can be regarded as the clinical predictors of restenosis after pereutaneous transluminal angioplasty. This paper aims to make a summarization of some kinds of clinical predictors which are related to inflammation.

ABSTRACT

Objective To study the effects of exercises training on the recovery of neurological function and the expression of matrix metalloproteinase-9 (MMP-9) in perihematomal brain tissue after intra cerebral hemorrhage (ICH) in rats. Methods Sixty-four male adult Sprague-Dawley rats were randomly divided into exercises group and control group. ICH model was induced by autobiood injection. The rats in exercises group were given balance, grasping and walking exercises every day. The rats in control group exercised freely in standard cages. Neurological function in both groups was measured at the 3rd, 7th, 14th and 21st d. All the rats were sacrificed and the concentration of MMP-9 was measured with immunohistochemical method and in situhybridization. Results In both groups neurological function scores was lowest at the 3rd d, were highest at the 21st d. There was no significant difference between two groups at the 3rd d, but at the 7th, 14th and 21st d the differences between two groups were significant ( P < 0.05 ). In exercises group, MMP-9 expressed weakly at the 3rd d, and peaked at the 7th d, then declined gradually. In control group, MMP-9 expression peaked at the 3rd d, then decreased slowly. MMP-9 protein expression was in accord with mRNA. MMP-9 expression in exer-cises group was significantly different from control group on the 3rd and 7th d (P < 0.05 ) , but not at the 14th and 21st d ( P > 0.05 ). Conclusions Early exercises can inhibit the expression of MMP-9 that could play a role in protecting neurons. Daily exercises can stimulates the expression of MMP-9 so as to have a positive role in midstage of disease. MMP-9 may be involved in tissue remodelling and vascular repairing, which prompt neu-rofunction recovery.

ABSTRACT

Background In recent years,some researches had been conducted on the pathologic changes of the secondary injury of perihematoma in animal experiments,but only a few studies had been done on the dynamic pathologic and ultrastructural changes of the perihematoma in ICH patients. The unique contribution of our study is to investigate the dynamic pathologic and ultrastructural changes of the perihematoma in ICH patients and provide significant insights into how the pathophysiology and ultrastructures changed after ICH.Methods The written informed consents were obtained from the ICH patients or their relatives. 30 patients (the supertentorial hemotoma volume>30 mi and the cerebellar hemotoma volume >10 mi) were divided into 8 groups according to the time passed after ICH:<6 h (6 patients), 6 ~ 12 h (7 patients), 12 ~24 h (5 patients), 24~48 h (3 patients), 48 ~72 h (3 patients), 3 ~4 days group (3 patients), 5 days group (2 patients) and 8 days group ( 1 patient) and subjected to craniotomy for hemotoma evacuation. During the operation for the hemotoma's evacuation, a small amount of tissues that must be removed, which located at 1 cm near the hematoma, were taken as experimental groups; And the same tissues of 7 patients (<12 h), which were far from the hemotoma on the operational way, were taken as control group. The pathologic and ultrastructral changes were observed.Results The tissues of the control group were almost normal while the damages of the tissues from the experimental groups were slight in <6 h groups, more severe after 6h and got to the maximum between 24 ~48 h , recovered gradually after 72 h, became similar to the 6 ~ 12 h group on 5 th day, got better on 8 th day and resembled the 6 h group.Conclusions The injury of the perihematoma occurred in early stage, reached the peak level between 24 and 48 hours after ICH; which was consistent to the clinical nervous functional deficits in the ICH patients.

ABSTRACT

In-stent restenosis after interventional treatment remains an unsolved and important clinical problem. The proliferation and migration of arterial smooth muscle cells are key events frequently followed by vascular restenosis after angioplasty and continuous inflammation may also be the other important factor for the restenosis after stenting. Study of the pathogenesis of restenosis may find some potentially novel therapeutic pathways for attenuating in-stent restenosis.

Objective To observe the expression of the Rho kinase in perihematomal cerebral tissue after experimental intracerebral hemorrhage(ICH) and the relationship between which and the neurological function deficit.Methods The ICH models rats were randomly divided into groups 1 d,3 d,5 d and 7 d after ICH.At the certain time point,the score of neurological function in each model was measured before killed.The expression of Rho kinase protein and mRNA were detected by immunohistochemistry and in situ hybridization.Results After ICH 1 d,3 d,5 d and 7 d groups,the scores of neurological function were 1.63,2.25,3.13 and 2.88,respectively;the number of Rho kinase protein positive cells in perihematomal cerebral tissue were 26.63,30.75,38.88 and 35.00,and the number of Rho kinase mRNA positive cells were 27.25,32.25,40.88 and 38.00,respectively.There were significantly differences among the groups 1 d,3 d and 5 d in the scores of neurological function and the expression of the Rho kinase protein and mRNA(P
